Many manufactures tend to dive into Human disease diagnostic field intensively at the first time, Bio TNS expands our horizon range from disease diagnostic not only human disease for example food poisoning but also aquatic animal disease for instance Red Sea bream Iridoviral disease to species detection of bacteria, virus and animal.

All of these contents developed following to take our own line on technical training and improving method for various Real-time PCR and reverse transcription(RT) PCR techniques, based on SYBR intercalator, TaqMan probe, and PNA (peptide nucleic acid).

Since that, although we are very small and may be non-famous, absence of brand name now, We catch the many chances to gain many experiences of researching and development various contents broad ranges and co-working achievements with Public institutions including Korea National Fishery Products Quality Management Service (NFGS) and Korea National Institute of Fisheries Science, also involved many National research projects reporting meaningful business performances. 2. Basic Principles

The majority of PCR methods rely on thermal cycling. Thermal cycling exposes reactants to repeated cycles of heating and cooling to permit different temperature-dependent reactions – specifically, DNA melting and enzyme-driven DNA replication. PCR employs two main reagents – primers (which are short single strand DNA fragments known as oligonucleotides that are a complementary sequence to the target DNA region) and a DNA polymerase. In the first step of PCR, the two strands of the DNA double helix are physically separated at a high temperature in a process called Nucleic acid denaturation. In the second step, the temperature is lowered and the primers bind to the complementary sequences of DNA. The two DNA strands then become templates for DNA polymerase to enzymatically assemble a new DNA strand from free nucleotides, the building blocks of DNA. As PCR progresses, the DNA generated is itself used as a template for replication, setting in motion a chain reaction in which the original DNA template is exponentially amplified.

Product Overview

The COVID-19 RT-qPCR PNA KIT is developed with reusable fluorescence hybridization probe real-time PCR technique using Peptide Nucleic Acid (PNA), sort of artificial nucleic acid which has more higher binding ability to its complement sequence DNA than which of DNA one, to in vitro reverse transcription of SARS-CoV-2 (also known as SARS-2019-nCoV) RNA followed with cDNA amplification and detection.

The kit targets three specific genomic regions of SARS-CoV-2, those are RNA dependent RNA Polymerase (RdRP) gene , Nucleocapsid (N). These are able to detect not only individual but also in the same time. And All the Positive results from detection indicative the presence of SARS-CoV-2, expectedly.

The PNA probes for the detection of SARS-CoV-2 are labeled FAM (N gene) and HEX(RdRP) fluorescent dyes. Target amplification results are shown target dependently different colors and able to analyze which target is detected easily.

PNA, the key components of this kit development technique, is sort of artificial analog of DNA which has neutrally charged peptide backbone. It has ultimate thermo stability with high specificity to the target nucleic acid. Additionally, It has also extreme enzyme stability, in other words, enzymes such as DNA polymerase or exonuclease do not recognize to polymerization or severing partial DNA-PNA double strand. This characteristic usefully contributes to struggle for preventing false positive results and the diagnostic grey zone as well.

This Kit also include Internal Positive control, which designed on Human acidic ribosomal protein (HuPO) gene, which was reported one of the standard to very fitting for normalization of RNA expression using PCR assay, for confirming correct RNA transcription to cDNA and all the PCR reaction procedure is do on right way with reliability of performance. The PNA probe for the Internal Positive control is labeled Cy5, so it is separately able to confirm the PCR reaction such as enzyme activity and PCR equipment productivity independent to SARS-CoV-2 results.
